Whole-mount Immunohistochemistry of Adult Zebrafish Retina for Advanced Imaging

[Abstract]  

Immunohistochemistry is a widely used technique to examine the expression and subcellular localization of proteins. This technique relies on the specificity of antibodies and requires adequate penetration of antibodies into tissues. The latter is especially challenging for thick specimens, such as embryos and other whole-mount preparations. Here we describe an improved method of immunohistochemistry for retinal whole-mount preparations. We report that a cocktail of three reagents, Triton X-100, Tween-20, and DMSO, in blocking and antibody dilution buffers strongly enhances immunolabeling in whole-mount retinas from adult zebrafish. In addition, we establish that in whole retinal tissues, a classic epitope retrieval method, based on citrate buffer, is effective for immunolabeling

[Abstract]  Murine tumor models have been critical to advances in our knowledge of tumor physiology and for the development of effective tumor therapies. Essential to these studies is the ability to both track tumor development and quantify tumor burden in vivo. For this purpose, the introduction of genes that confer tumors with bioluminescent properties has been a critical advance for oncologic studies in rodents. Methods of introducing bioluminescent genes, such as firefly luciferase, by viral transduction has allowed for the production of tumor cell lines that can be followed in vivo longitudinally over long periods of time. Here we describe methods for the production of stable luciferase expressing tumor cell lines by lentiviral transduction. [Abstract]  Corn, or maize, is a major cereal crop and model monocot. Two of its benefits are ease of pollination and tractable genetics. For a comparison of development between maize and the dicot model, Arabidopsis, see Lunde and Hake, 2005. For an example of how this protocol might be used for a double mutant analysis see Lunde and Hake, 2009. Here, we provide a protocol for easy DNA extraction without phenol or chloroform. Therefore, this protocol is suitable for use in schools or laboratories that lack fume hoods.
