Fluorescent Labeling of Rat-tail Collagen for 3D Fluorescence Imaging

[Abstract]  Rat tail collagen solutions have been used as polymerizable in vitro three-dimensional (3D) extracellular matrix (ECM) gels for single and collective cell migration assays as well as spheroid formation. These 3D hydrogels are a relatively inexpensive, simple to use model system that can mimic the in vivo physical characteristics of numerous tissues within the body, namely the skin. While confocal imaging techniques such as fluorescence reflection and two-photon microscopy are able to visualize collagen fibrils during 3D imaging without fluorescence, other imaging modalities require direct conjugation of fluorescent dyes to collagen. [Abstract]  Angiogenesis defines the process of formation of new vascular structures form existing blood vessels, involved during development, repair processes like wound healing but also linked to pathological changes. During angiogenic processes, endothelial cells build a vascular network and recruit perivascular cells to form mature, stable vessels. Endothelial cells and perivascular cells secret and assemble a vascular basement membrane and interact via close cell-cell contacts. To mimic these processes in vitro we have developed a versatile three-dimensional culture system where perivascular cells (PVC) are co-cultured with human umbilical cord vascular endothelial cells (HUVEC) in a collagen type I gel. [Abstract]  Cancer tissue is composed of cancer cells and a large number of stromal cells including fibroblasts. In order to understand the relationship between fibroblasts and cancer cells during invasion of the stroma, 3D gel invasion assay is useful. Most tumors are associated with a biologically active type of fibroblasts known as cancer-associated fibroblasts (CAFs), which promote the invasion of cancer cells. Here, we describe the method of imaging the invasion by fluorescently labeled CAFs and gastric cancer cells in gels containing extracellular matrix. For two-color fluorescence labeling of living cells, long-chain dialkylcarbocyanines, DiO and DiI were used.
